Gluconeogenesis (GNG) is a metabolic pathway that results in the biosynthesis of glucose from certain non-carbohydrate carbon substrates. It is a ubiquitous process, present in plants, animals, fungi, bacteria, and other microorganisms. In vertebrates, gluconeogenesis occurs mainly in the liver and, to a lesser extent, in the cortex of the kidneys. It is one of two primary mechanisms – the other being degradation of glycogen (glycogenolysis) – used by humans and many other animals to maintain blood sugar levels, avoiding low levels (hypoglycemia). In ruminants, because dietary carbohydrates tend to be metabolized by rumen organisms, gluconeogenesis occurs regardless of fasting, low-carbohydrate diets, exercise, etc. In many other animals, the process occurs during periods of fasting, starvation, low-carbohydrate diets, or intense exercise. In humans, substrates for gluconeogenesis may come from any non-carbohydrate sources that can be converted to pyruvate or intermediates of glycolysis (see figure). From the breakdown of proteins, these substrates include glucogenic amino acids (although not ketogenic amino acids); from breakdown of lipids (such as triglycerides), they include glycerol, odd-chain fatty acids (although not even-chain fatty acids, see below); and from other parts of metabolism that includes lactate from the Cori cycle. Solvent extraction is used extensively in separation and purification processes. In its simplest form a reaction is performed in an organic solvent and unwanted by-products are removed by extraction into water at a particular pH. A metal ion may be extracted from an aqueous phase into an organic phase in which the salt is not soluble, by adding a ligand. The ligand, La−, forms a complex with the metal ion, Mb+, [MLx](b−ax)+ which has a strongly hydrophobic outer surface. Currently extraction into kerosene is preferred, using a ligand such as tri-n-butyl phosphate, TBP. In the PUREX process, which is commonly used in nuclear reprocessing, uranium(VI) is extracted from strong nitric acid as the electrically neutral complex [UO2(TBP)2(NO3)2]. The strong nitric acid provides a high concentration of nitrate ions which pushes the equilibrium in favour of the weak nitrato complex. Uranium is recovered by back-extraction (stripping) into weak nitric acid. Plutonium(IV) forms a similar complex, [PuO2(TBP)2(NO3)2] and the plutonium in this complex can be reduced to separate it from uranium.

=== Mayor of Nuevo Cuscatlán === In 2011, Bukele announced that he would enter politics as a member of the FMLN to break out of "his comfort zone" ("su zona de confort") as a businessman. Officially joining the party in 2012, he campaigned for the mayoralty of Nuevo Cuscatlán, a municipality in the department of La Libertad, part of the San Salvador metropolitan area. Bukele's campaign was supported by the Democratic Change party. He was elected mayor of Nuevo Cuscatlán in March 2012 with 51.67 percent of the vote, defeating primary challenger Tomás Rodríguez of the Nationalist Republican Alliance (ARENA). Bukele took office on 1 May 2012 as the country's youngest mayor. Bukele created a scholarship program for youths in the municipality, donating his $2,000 salary to fund the program. In August 2014, Bukele launched Sphere PM, a project that launched a high-altitude balloon to an altitude of 100,000 feet (30,000 m) and took pictures of El Salvador. He stated that Sphere PM's goal was to promote education in science and technology to dissuade the municipality's youth from crime. Bukele spoke at the United Nations headquarters about projects he had undertaken as mayor of Nuevo Cuscatlán as part of the November 2014 World Cities Day. In January 2015, he inaugurated a $1.7 million boulevard connecting Nuevo Cuscatlán with Huizúcar and Antiguo Cuscatlán. Bukele did much of his mayoral work with funding from ALBA Petróleos, owned by the Venezuelan oil company PDVSA.

=== Erroneous and incomplete discoveries === In 1914, Stefan Meyer, Viktor F. Hess, and Friedrich Paneth (working in Vienna) made measurements of alpha radiation from various substances, including 227Ac. They observed the possibility of a minor alpha branch of this nuclide, though follow-up work could not be done due to the outbreak of World War I. Their observations were not precise and sure enough for them to announce the discovery of element 87, though it is likely that they did indeed observe the decay of 227Ac to 223Fr. Soviet chemist Dmitry Dobroserdov was the first scientist to claim to have found eka-caesium, or francium. In 1925, he observed weak radioactivity in a sample of potassium, another alkali metal, and incorrectly concluded that eka-caesium was contaminating the sample (the radioactivity from the sample was from the naturally occurring potassium radioisotope, potassium-40). He then published a thesis on his predictions of the properties of eka-caesium, in which he named the element russium after his home country. Shortly thereafter, Dobroserdov began to focus on his teaching career at the Polytechnic Institute of Odesa, and he did not pursue the element further. The following year, English chemists Gerald J. F. Druce and Frederick H. Loring analyzed X-ray photographs of manganese(II) sulfate. They observed spectral lines which they presumed to be of eka-caesium. They announced their discovery of element 87 and proposed the name alkalinium, as it would be the heaviest alkali metal.
The two main carbon-containing fragments are combined by the reaction of an aryl lithium with a trimethylsilyl-protected gluconolactone. The trimethylsilyl groups are then removed by treatment with methanesulfonic acid in methanol. This gives an intermediate with an unwanted methoxy group at the anomeric centre, which is removed by reaction with triethylsilane in the presence of boron trifluoride etherate. This route, as well as others developed for the manufacture of the drug, have been reviewed.

=== Melanoma === Tumour-associated macrophages can be attracted by cytokines such as TGF-β and IL-10 produced by tumours in melanoma. These cytokines can also trigger the M2 polarisation of the infiltrated tumour-associated macrophages. Infiltrated macrophages that acquire the M2 phenotype are anti-inflammatory. They facilitate tumour growth and proliferation via immunosuppression, further cytokine production, and tumour angiogenesis. Tumour-associated macrophages also promote the invasion of cancer cells via the production of miRNA exosomes.
== Tissue development == Haptotaxis plays a role in organizing cells to form tissues and specific regions of those tissues. Fibronectin and laminin both play a role in adrenocyte mutation into distinctive distribution in the adrenal gland. The adrenocytes migrate centripetally as they mature towards the medulla of the adrenal gland, and this movement may be a result of haptotactic forces mediated by fibronectin and laminin. In nerve cells, axonal growth is mediated by nerve growth factor in a haptotactic manner, where the axon of nerve cells grows along the gradient. This information could be used to possibly develop methods to promote nerve regeneration in patients that have nerve damage. Another regenerative strategy is the use of mesenchymal stem cells, which can differentiate into different kinds of connective tissue in the wound healing process. The haptotaxis is mediated by fibronectin, vitronectin, and type I collagen. A recent study has tentatively proposed the idea that the structures on cells responsible for sensing the membrane protein gradients are attenuated filopodia. Also, the more amount of filopodia present on the leading edge of the migrating cell, the more responsive the cell is to the haptotactic gradient. This is important because there is the possibility that all motile cells that display filopodia may be responding to haptotactic gradients. Further research is required in the subject, but it is clear that more and more kinds of cell undergo haptotaxis than originally believed.

In this process, a ligand is immobilized on the dextran surface of the SPR crystal. Through a microflow system, a solution with the analyte is injected over the ligand-covered surface. The binding of the analyte to the ligand causes an increase in the SPR signal (expressed in response units, RU). Following the association time, a solution without the analyte (typically a buffer) is introduced into the microfluidics to initiate the dissociation of the bound complex between the ligand and analyte. As the analyte dissociates from the ligand, the SPR signal decreases. From these association ('on rate', ka) and dissociation rates ('off rate', kd), the equilibrium dissociation constant ('binding constant', KD) can be calculated. The detected SPR signal is a consequence of the electromagnetic 'coupling' of the incident light with the surface plasmon of the gold layer. This interaction is particularly sensitive to the characteristics of the layer at the gold–solution interface, which is usually just a few nanometers thick. When substances bind to the surface, it alters the way light is reflected, causing a change in the reflection angle, which can be measured as a signal in SPR experiments.
