Thymosin alpha-1 is a short peptide of 28 amino acid residues first described in the 1970s as a component of thymic extracts. Its N-terminal residue carries an acetyl group, and the sequence is highly conserved across mammalian species. The peptide is not encoded as a standalone gene product; it is released by proteolytic cleavage from the N-terminus of prothymosin alpha, a larger acidic nuclear protein. That precursor relationship places it within a broader family of thymic and immune-associated peptides that have been studied for decades.
The activity of this peptide is generally described as immunomodulatory rather than directly antimicrobial. Experimental work links it to signaling through certain Toll-like receptors on dendritic cells and to downstream maturation of antigen-presenting cells. Reported effects include expansion of T cell subsets, shifts in cytokine profiles, and increased natural killer cell activity. These observations come largely from cell culture and animal models, and the precise receptor-level events in humans remain incompletely characterized.

Within the immune system, the peptide acts on several cell types rather than a single target. Reported activities include promotion of T-cell maturation, enhancement of natural killer cell activity, and modulation of cytokine production by dendritic cells and macrophages. Some of these effects appear to operate through toll-like receptor signaling, though the precise receptor-level mechanism remains debated. Whether the observed immune changes translate into clinical benefit is a separate question and depends on the indication studied.
The peptide was described in the 1970s as a component of thymic extracts, and early research focused on restoring immune function in immunodeficiency states. A synthetic version entered clinical development in the 1980s and is approved as a drug in several countries for conditions such as chronic hepatitis B and certain immunodeficiencies. Approval status varies widely by jurisdiction, and in the United States it is not an approved therapeutic. Regulatory and clinical positions differ, so statements about efficacy should be tied to specific indications and studies.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids, corresponding to the N-terminal fragment of prothymosin alpha. Its sequence begins with acetylation at the N-terminus, a modification that affects stability and receptor interaction. The peptide is acidic, with a calculated isoelectric point near 4.2, and carries no disulfide bonds, so its secondary structure is largely flexible in solution. Molecular mass is approximately 3108 daltons. The native form was first isolated from bovine thymus tissue, while pharmaceutical material is produced by solid-phase peptide synthesis.
Stability depends on temperature, pH, and the number of freeze-thaw events the sample has experienced. Freeze-dried material is commonly held at -20 °C or colder, while reconstituted liquid is kept cold and used within a short window. Extreme pH and prolonged light exposure can promote deamidation, oxidation, or aggregation, particularly at asparagine and methionine positions. Adsorption to container walls can lower the measured concentration of a dilute solution even when the peptide molecules themselves remain intact.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, which separates the target peptide from truncated or chemically modified byproducts. Mass spectrometry confirms the expected molecular mass and can indicate acetylation state or sequence errors. Amino acid analysis and peptide mapping supply complementary sequence-level information, while endotoxin testing is relevant for preparations intended for cell or animal work. Purity figures reported by suppliers refer to the method used and are not directly comparable across laboratories unless conditions are stated.
Several factors accelerate degradation: alkaline pH, elevated temperature, exposure to oxidants, and the presence of residual moisture. Deamidation of asparagine residues and oxidation of methionine are the most commonly reported degradation routes. Because the peptide lacks disulfide bonds, it does not undergo the thiol-related aggregation seen in some other biologics, but physical aggregation can still occur at high concentration. Stability data are product-specific, and extrapolating shelf life between formulations is not reliable.
Lyophilized thymosin alpha-1 is generally stored at or below minus twenty degrees Celsius, protected from moisture and light. Short-term handling at ambient temperature is possible for dry powder, but reconstituted solutions degrade faster and are usually kept at two to eight degrees Celsius with a defined expiry of days rather than weeks. Repeated freeze-thaw cycles should be avoided because they promote aggregation and loss of potency. Exact limits depend on the formulation and should follow the supplier's documentation.
=== Plants === In plants, ADH catalyses the same reaction as in yeast and bacteria to ensure that there is a constant supply of NAD+. Maize has two versions of ADH – ADH1 and ADH2, Arabidopsis thaliana contains only one ADH gene. The structure of Arabidopsis ADH is 47%-conserved, relative to ADH from horse liver. Structurally and functionally important residues, such as the seven residues that provide ligands for the catalytic and noncatalytic zinc atoms, however, are conserved, suggesting that the enzymes have a similar structure. ADH is constitutively expressed at low levels in the roots of young plants grown on agar. If the roots lack oxygen, the expression of ADH increases significantly. Its expression is also increased in response to dehydration, to low temperatures, and to abscisic acid, and it plays an important role in fruit ripening, seedlings development, and pollen development. Differences in the sequences of ADH in different species have been used to create phylogenies showing how closely related different species of plants are. It is an ideal gene to use due to its convenient size (2–3 kb in length with a ≈1000 nucleotide coding sequence) and low copy number.
In the early modern period, successor states to the Adal Sultanate and Ajuran Sultanate began to flourish in Somalia. These included the Hiraab Imamate, the Isaaq Sultanate led by the Guled dynasty, the Habr Yunis Sultanate led by the Ainanshe dynasty, the Sultanate of the Geledi (Gobroon dynasty), the Majeerteen Sultanate (Migiurtinia), and the Sultanate of Hobyo (Obbia). They continued the tradition of castle-building and seaborne trade established by previous Somali empires. Sultan Yusuf Mahamud Ibrahim, the third Sultan of the House of Gobroon, started the golden age of the Gobroon Dynasty. His army came out victorious during the Bardheere Jihad, which restored stability in the region and revitalised the East African ivory trade. He also had cordial relations and received gifts from the rulers of neighbouring and distant kingdoms such as the Omani, Witu and Yemeni Sultans. Sultan Ibrahim's son Ahmed Yusuf succeeded him as one of the most important figures in 19th-century East Africa, receiving tribute from Omani governors and creating alliances with important Muslim families on the East African coast.
=== 2026 Data breach and class action === On January 28, 2026, Panera Bread was hit by a cyber attack, along with other companies including Bumble and Match. The company revealed that contact information was the data involved. Later reporting noted that the aggressor was ShinyHunters, that they stole information on around 5 million people (14 million records), and subsequently leaked this information to the dark web. Two class action lawsuits were filed against the company shortly after, citing Panera Bread's failure to properly secure the personal information that was stolen.
== Origin == The first symptoms of apitoxin (bee venom), that are now thought to be caused by apamin, were described back in 1936 by Hahn and Leditschke. Apamin was first isolated by Habermann in 1965 from Apis mellifera, the Western honey bee. Apamin was named after this bee. Bee venom contains many other compounds, like histamine, phospholipase A2, hyaluronidase, MCD peptide, and the main active component melittin. Apamin was separated from the other compounds by gel filtration and ion exchange chromatography.
=== Occurrence in animals === Numerous arthropods (Arthropoda) contain cyanogenic (hydrogen cyanide-releasing) nitrile compounds, including centipedes (Chilopoda), bipedes (Diplopoda), beetles (Hemiptera), beetles (Coleoptera), and butterflies (Lepidoptera). The gooseberry moth (Abraxas grossulariata) contains the nitrile-bearing glycoside sarmentosin, which likely functions in defense. Sarmentosin is also present in several species of the genus Parnassius. Several species of glass-winged bug (including Jadera haematoloma) contain cyanolipids or cardiospermine, which they may acquire from their host plants through sequestration of toxins, i.e., uptake and storage. Six-spotted damselflies are butterflies capable of both sequestering the cyanogenic glycosides linamarin and lotaustralin from their host plants and synthesizing them de novo. Other species of the same genus (Zygaena), such as the Marsh Hornwort, also contain cyanogenic glycosides. The defensive secretion of the centipede Himantarium gabrielis contains benzoyl cyanide, phenylacetonitrile, mandelonitrile (benzaldehyde cyanohydrin), and mandelonitrile benzoate. Phenylacetonitrile also functions as a hormone in the desert locust (Schistocerca gregaria). In various tapeworms (Polydesmida), the defensive secretion likewise contains benzoyl cyanide. The mite species Oribatula tibialis (order horn mite, Oribatida) contains mandelonitrile hexanoate. Hydrogen cyanide also occurs in arthropods as a degradation product of cyanogenic compounds.

=== Cancer === Point mutations in multiple tumor suppressor proteins cause cancer. For instance, point mutations in Adenomatous Polyposis Coli promote tumorigenesis. A novel assay, Fast parallel proteolysis (FASTpp), might help swift screening of specific stability defects in individual cancer patients.
In chemical analysis, capillary electrochromatography (CEC) is a chromatographic technique in which the mobile phase is driven through the chromatographic bed by electro-osmosis. Capillary electrochromatography is a combination of two analytical techniques, high-performance liquid chromatography and capillary electrophoresis. Capillary electrophoresis aims to separate analytes on the basis of their mass-to-charge ratio by passing a high voltage across ends of a capillary tube, which is filled with the analyte. High-performance liquid chromatography separates analytes by passing them, under high pressure, through a column filled with stationary phase. The interactions between the analytes and the stationary phase and mobile phase lead to the separation of the analytes. In capillary electrochromatography capillaries, packed with HPLC stationary phase, are subjected to a high voltage. Separation is achieved by electrophoretic migration of solutes and differential partitioning.
Thomas Bellchambers (born 9 July 1989) is a former professional Australian rules footballer who played for the Essendon Football Club in the Australian Football League (AFL). He is a current professional boxer. After playing for the Tasmanian Devils in the TAC Cup, he was drafted with pick 8 in the 2007 pre-season draft. He made his debut against Hawthorn in Round 11 of the 2008 AFL season. He announced his retirement from the AFL on the 15th of September 2020. Bellchambers, along with 33 other Essendon players, was found guilty of using a banned performance-enhancing substance, thymosin beta-4, as part of Essendon's sports supplements program during the 2012 season. He and his team-mates were initially found not guilty in March 2015 by the AFL Anti-Doping Tribunal, but a guilty verdict was returned in January 2016 after an appeal by the World Anti-Doping Agency. He was suspended for two years which, with backdating, ended in November 2016; as a result, he served approximately fourteen months of his suspension and missed the entire 2016 AFL season. He announced his retirement on 15 September 2020, after 136 games. On 24/05/23 Bellchambers made his professional boxing debut Vs Cameron Mooney in Melbourne, Bellchambers won by KO in the 3rd round. The fight was in the heavyweight division. Bellchambers professional boxing record is 1-0. (1 by KO)

It is usually classified as an immunomodulatory peptide rather than a classical hormone. It derives from the larger protein prothymosin alpha and acts mainly on immune cells. The thymosin label covers a group of distinct peptides, so the naming can be misleading.
